Abstract(Please copy/paste the abstract send to the congress) : The main goal of periodontal treatment is to achieve regeneration with reconstruction of all the suitable components including periodontal ligament, alveolar bone, root cementum and connective tissue. The goal of this study was to assess the effect of purmorphamine (PMA) along with Collagen-Hydroxyapatite scaffold in inducing osteogenesis of endometrial stem cells (hEnSCs) in 2D and 3D culture media. The cells were cultured in an osteogenic differentiating media containing PMA in tissue culture plate (TCP as 2D group) and Collagen-Hydroxyapatite Scaffold (as 3D group) for 21 days. Structure of Collagen-Hydroxyapatite scaffold, cell morphology and cell attachment were analyzed with SEM and DAPI staining. The viability of cells on scaffold and osteoblast cells markers were assayed with MTT assay and real time PCR respectively. The release of ALP in 2 groups was compared. Cell viability is enhanced in Collagen-Hydroxyapatite scaffold after 7 days. SEM and DAPI staining show that cells are in good integration with structure of scaffold. Also, qRT-PCR of differentiated cells showed that COL-I, Osteocalcin (Bglap), bone sialoprotein (ibsp) and Runx-2 are expressed after 21days in 2D and 3D groups, in 3D group the expression of these markers were higher than 2D group. Under the effect of PMA the release of alkaline phosphatase increased incrementally in 3D group rather than 2D group during the 21 days of exposing the induction media. Based on our findings, Collagen-Hydroxyapatite Scaffold with PMA has the potential role in osteogenic differentiation of EnSCs and can be used to induce osteogenesis in bone defects.
